Abstract

Atherosclerosis is defined as a slow process of inflammation fuelled by deposition of lipids. The pathogenesis of atherosclerotic changes is associated with low High Density Lipid (HDL) and the mechanism is believed to be due to the low level of paraoxonase 1 (PON-1) located on it. Administration of chlorpyrifos (CPF) is known to reduce PON-1 level. This study is aimed to demonstrate atherosclerotic changes in rats’ aortas following exposure to CPF. Eighteen male Sprague-Dawley rats weighing 150-200 g were grouped into control, vehicle and CPF group. Control rats were not given any injection. Vehicle group was administered with 3% Dimethyl sulphoxide and 97% v/v soy oil while CPF exposed rats received 18 mg/kg CPF dissolved in vehicle with a volume of 0.7 ml/kg body weight. The rats were injected subcutaneously every other day for 6 months. The arch of aorta from each rat was processed for SEM. No obvious structural change observed in the aortas of control and vehicle groups. In aortas of CPF exposed group, there were disruptions of the intimal layer, numerous endothelial gaps and areas of deendothelialization were observed. This study demonstrated that CPF administration had caused vascular damage and supports the possible mechanism of low level PON-1 in the pathogenesis of atherosclerosis.
