Abstract

Ethanolic nano lipid vesicles are also known as ethosomes. They are novel elastic vesicles used as non-invasive drug delivery carriers for transdermal applications. This ethosomal system is composed of phospholipid, ethanol and water. The purpose of this research was to develop and characterize an ethosomal formulation of raloxifene HCl to deliver the drug into systemic circulation through the transdermal route. This was aimed to minimize the dose of the drug and achieve the maximum bioavailability in order to reduce the side effects associated with its repeated dose regimen. The drug loaded ethosomal formulation was prepared by the rotary evaporation method. The size, zeta potential and polydispersity index (PDI) of the formulation were measured. The drug entrapment efficiency of the vesicles was determined by an ultracentrifugation method. Ex vivo skin permeation study was conducted to measure transdermal flux (J) by Franz diffusion cell with rat skin as a barrier membrane. The advanced structural and skin penetration characterizations were carried out by force emission scanning electron microscope (FESEM), transmission electron microscope (TEM). Confocal laser scanning microscopy (CLSM) study was performed to view the permeated nanovesicles in the skin layers. The size, charge, PDI and ex vivo transdermal flux (J) of the developed formulation showed favorable values required for the stability and efficient transdermal drug delivery. FESM and TEM study confirmed nano size and structural morphology of the vesicles. CLSM microscopic studies proved an enhanced drug penetration into the different skin layers compared to the conventional liposomes and drug solution in ethanolic phosphate buffer. The developed nano lipid vesicles (ethosomes) might be promising carriers for transdermal drug delivery of raloxifene HCl.
