Abstract

Leaf anatomical and micromorphological characteristics of two species from the Fabaceae family, collected in Kuantan, Pahang, were examined to identify features useful for distinguishing similarities and differences at the genus and species levels. The method used in this study was plant histology method including cross section using sliding microscope, leaf epidermal peeling and scanning electron microscopy (SEM) for leaf micromorphology analysis. The findings revealed both shared and distinct anatomical traits that could serve as valuable tools for species identification, differentiation, and classification. Common characteristics observed in both species included the presence of cuticle layers, paracytic stomata, mucilage cells, and sclerenchyma tissue. However, several distinctive traits were also identified. These include the presence of trichomes, differences in the outline of the leaf margin, and variations in the anticlinal wall patterns of the adaxial and abaxial epidermis as well as type of waxes. Senna alata was the only species found to possess trichomes on both surfaces of the leaf. Three types of trichomes were recorded in Senna alata: simple unicellular trichomes (short, pointed, with echinate ornamentation) on the adaxial surface, peltate and papillae trichomes on the abaxial surface. Regarding leaf margin outline, Tamarindus indica displayed a straight margin with a rounded tip, while Senna alata showed a straight margin with 90° curved, tapered end. Additionally, the anticlinal wall patterns varied between the species, which is Tamarindus indica exhibited straight to wavy walls, while Senna alata showed wavy to sinuous patterns. Five types of waxes also been report in this study such as flakes and film layers were observed in Senna alata, while Tamarindus indica exhibited granular, rod-shaped, and verrucate wax forms. In conclusion, this anatomical study highlights the presence of distinct and species-specific anatomical characteristics, supporting their value in the taxonomic identification and classification of plant species, particularly within the Fabaceae family.
