Abstract

Portable, low-cost platform and sensing system for detection and quantification of analytes are in high demand for various environmental monitoring applications especially for field work and humanitarian use. Quantifying analytes and relevant water-quality parameters in the field requires a device that can perform measurements with high sensitivity, stability, and fast response time. A portable device capable of pathogen and ion detection and quantification is useful for evaluating water quality in situ. Our electrochemical sensor technology can provide water-quality assessment, especially for detection of pathogenic Escherichia coli (E. coli) O157:H7 and relevant ions (Ca2+, Mg2+, HCO3-/CO32-, NH4+, and Cl-). We characterized and optimized reduced graphene and conductive polymer deposition methods on glassy-carbon (GCE) and platinum (Pt) electrodes respectively, for biomolecule immobilization. An electrochemical immunosensor for E. coli detection was fabricated through covalent immobilization of anti-E. coli O157:H7 onto reduced graphene-based electrodes. The initial results based on cyclic voltammetry (CV) studies show promising functionality of the graphene-based immunosensor to detect low concentration of E. coli O157:H7 with a wide detection range. Functionality of conductive-polymer based sensor was tested by detecting and monitoring pH, Ca2+, Mg2+, HCO3-/CO32-, NH4+, and Cl- ions. The goal is to develop a versatile device with multiple sensors that are useful for water quality assessment and monitoring; the sensors are further integrated in a portable water system platform for field work and humanitarian use.
