Abstract

Mangosteen (Garcinia mangostana var. mangostana) is one of the common tropical fruits in South East Asia. The value and economic potential of mangosteen have been recognised by local and international markets due to its fleshy part of the pulp that can be eaten fresh as well as medicinal properties including anti-inflammatory and anti-oxidative characteristics. Mangosteen grows best in fertile soil with slightly acidic conditions and can only be found in cultivated farms especially in few countries including Thailand, Malaysia, Philippines, and Indonesia. In order to improve both quality and quantity of mangosteen cultivation, several Garcinia species have been identified as possible ancestors of mangosteen which can be used in breeding program. Currently, the genome size on these possible ancestors are still unavailable. This study aims to determine the genome size of two Garcinia species of possible mangosteen ancestors; G. opaca and G. penangiana using flow cytometry (FCM) analysis. By using soybean or Glycine max cv. Polanka (2C=2.5pg) as an external reference standard, it was found that the genome size of G. opaca and G. penangiana to be 3.34±0.05 pg and 4.77±0.31* pg respectively. Recent study proved that the genome size of mangosteen to be 6.00±0.17 pg which was higher than the genome size of both G. opaca and G. penangiana. The findings indicate that interspecific variation which resulted from hybridization process between G. opaca and G. penangiana may occur in the genome of mangosteen. In conclusion, cytological data such as genome size for Garcinia species can be utilised for further study to determine the origin of mangosteen.
